Modern laboratory methods for detecting antibiotics

За последние десятилетия был разработан ряд современных лабораторных методов, предлагающих высокую чувствительность, специфичность и скорость.
1. Микробиологические методыЭти методы основаны на способности антибиотиков ингибировать рост чувствительных микроорганизмов. Они являются классическими и часто используются в качестве референтных.
- Диффузия в агар (методы дисков и лунок): Антибиотик диффундирует из диска или лунки в питательную среду, засеянную бактериями. Зона ингибирования роста вокруг диска/лунки указывает на присутствие и активность антибиотика.
- Определение минимальной ингибирующей концентрации (МИК): Позволяет количественно определить наименьшую концентрацию антибиотика, которая полностью подавляет видимый рост бактерий. Используются методы разведения в бульоне или на агаре.
- Рецепторно-микробиологические методы: Используют специфические рецепторы бактериальных клеток для связывания антибиотиков, что затем приводит к ингибированию роста.
Эти методы основаны на высокоспецифичном взаимодействии антигена (антибиотика) с соответствующим антителом. Они характеризуются высокой скоростью и возможностью автоматизации.
- Иммуноферментный анализ (ELISA): Широко используемый метод для качественного и количественного определения антибиотиков. Существуют прямые, непрямые и конкурентные варианты ELISA, где связывание антибиотика с антителом детектируется посредством ферментативной реакции, приводящей к изменению цвета.
- Иммунохроматографический анализ (тест-полоски, Lateral Flow Assays): Экспресс-методы, которые обеспечивают быстрое обнаружение антибиотиков без сложного оборудования, идеальны для скрининга на месте. Результат обычно проявляется в виде изменения цвета на тест-полоске.
- Иммуносенсоры: Современные устройства, объединяющие биохимическое распознавание с физическим преобразователем для регистрации специфического взаимодействия антиген-антитело.
Эти методы направлены на обнаружение генетических маркеров устойчивости к антибиотикам или генов, кодирующих ферменты, деактивирующие антибиотики, а также на прямую идентификацию самих микроорганизмов.
- Polymerase chain reaction (PCR) and real-time PCR (qPCR): Used to amplify and detect specific DNA fragments associated with antibiotic resistance genes or the presence of antibiotic-producing bacteria. qPCR allows for the quantitative assessment of the target DNA content.
- Next-generation sequencing (NGS): Allows comprehensive analysis of the entire genome or specific regions of microorganisms to identify all potential antibiotic resistance genes.
These methods are based on the separation of the components of the mixture and their subsequent identification and quantitative determination.
- High-performance liquid chromatography (HPLC): Widely used for the separation, identification, and quantification of antibiotics in complex matrices. Often combined with various detectors (UV/visible, fluorescence).
- Liquid chromatography with mass spectrometry (LC-MS/MS): the "gold standard" for accurate and sensitive detection of antibiotics. It allows not only separation but also identification of compounds by their mass and fragments, ensuring high specificity and low detection limits even in highly complex samples.
- Gas Chromatography with Mass Spectrometry (GC-MS/MS): Less common for antibiotics that require derivatization to increase volatility, but can be used for some classes.
The choice of antibiotic detection method depends on the sample type, required sensitivity, specificity, analysis speed, and available equipment. Modern laboratory methods, especially LC-MS/MS, offer unprecedented opportunities for accurate and reliable antibiotic monitoring, contributing to food safety, environmental monitoring, and the effective fight against antibiotic resistance.
